【中文摘要】 本研究通過電鏡觀察與酶解分析確定香菇(Lentinula edodes)菌絲細胞壁組成.并進行了雙核菌絲原生質(zhì)體的分離和培養(yǎng).研究發(fā)現(xiàn):酶解過程中,菌絲原生質(zhì)體一般只從菌絲頂端等新生部位釋放出來.合適的滲透劑、菌齡和酶解溫度是原生質(zhì)體得率的重要保證;原生質(zhì)體再生菌絲過程是一個間歇性的依賴于原生質(zhì)團涌動流向的過程,再生菌絲的形成經(jīng)歷了原生質(zhì)體形成突起-啞鈴狀-分枝狀等不同形態(tài)類型的再生階段;再生菌絲體在形態(tài)上沒有觀察到鎖狀聯(lián)合,栽培后未能結實,初步判斷為單核菌株.

【英文摘要】 The composition of Lentinuta edodes hypha cell wall is determined by electron microscope observation and enzyme digestion analysis. And the protoplast isolation from dikaryotic mycelium and culture are also carried out in this study. The results showed that the protoplast was released generally from new growth part such as hypha tips through enzymolysis and the appropriate osmotic stabilizer, the strain age and the temperature during enzymolysis were all the important factors for high protoplast yield, that the procedure of protolast hypha regeneration was a procedure of intermittence depending on the direction of protoplasmatic mass movement and the formation of protoplast regenerated hypha experienced protruding-shape stage, dumbbell shape stage and branchy shape stage, and that no clamp connection was observed during mycelium growth period, the fruitbody could not be formed from the protoplast strain and the strain regenerated from protoplast might be the monokaryotic strain.
